Öğe Application of Half-Sandwich Metal-Phosphinite Compounds to Biological Activities: Determine the energies of the HOMO and LUMO levels(Wiley Online Library, 2022) Meriç, Nermin; Rafikova, Khadichakhan; Zazybin, Alexey; Güzel, Remziye; Kayan, Cezmi; Karakaş, Duygu Elma; Dündar, Abdurrahman; ISLAM, Sholpan; Okumuş, Veysi; Aydemir, MuratMononuclear transition metal complexes 1-(furan-2-yl)ethyldiphenyl[dichloro(η6-p-cymene)ruthenium(II)]phosphinite, (2), 1-(furan-2-yl)ethyldiphenyl[dichloro(η6-benzene) ruthenium(II)] phosphinite (3), 1-(furan-2-yl)ethyldipheny[chloro(η4-1,5-cyclooctadiene)rhodium(I)]phosphinite (4), 1-(furan-2-yl)ethyldiphenyl[dichloro (η5pentamethylcyclopentadienyl)iridium (III)] phosphinite (5) were synthesized and characterized by microanalysis, infrared, MS, and NMR spectroscopy. The biological activities of the complexes were also tested. Compounds 2 and 5 were the best complexes at DPPH radical scavenging and reducing power activity at 73.27 % and 0.41 at 200 μg/mL, respectively. The highest antimicrobial activity exhibited by complex 3 as 14 mm inhibition zone against S. aureus. All of the complexes have cleaved the DNA from the double-strand and exhibited three bands on gel electrophoresis. Moreover, cyclic voltammetry studies of the phosphinite complexes were carried out to determine the energies of the HOMO and LUMO levels as well as to estimate their electrochemical and some electronic properties.Öğe Assessment Of The Antimicrobial, Antioxidant And Cytotoxic Activities Of The Wild Edible Mushroom Pleurotus Eryngii From Hakkari, Turkey(2017) Acay, Hilal; Dündar, Abdurrahman; Keskin, Cumali; Kaçar, SemraAbstract: Mushroom is an important source of natural compounds with acknowledged bioactivity. Pleurotus eryngii, in particular, is widely recognized for its organoleptic quality and favorable healthy effects, being commercially produced in great extent. The present study is just for evaluating the antioxidant capacity (scavenging DPPH radicals, reducing power, Total antioxidant activity by β-carotene-linoleic acid metod, Chelating effect on ferrous ions, superoxide anion radical scavenging activity ), antimicrobial effects (by using the disc diffusion method) and insufficiently studied cytotoxic activity (using PC-3 cell lines) of Pleurotus eryngii by using various solvent systems. For this reason; obtained extracts of the mushroom applying hexane, ethylacetate and methanol respectively were tested with in its biologic activities. As a result, The highest total antioxidant activity by β -carotene-linoleic acid metod was identified in methanol extract (63.62 mg /ml). The highest DPPH scavenging activity was obtained from ethyl acetate extract with 88.81 mg/ml. EDTA is used as standard in methal chelating activity. The highest metal chelating activity of Pleurotus eryngii identified in hexane (88,1 mg /ml) and ethylacetate (88.84 mg /ml) extracts. Additionally, in most of studied tests antioxidant activity of the mushroom increased with increasing concentration. On the other hand, the highest antimicrobial activity was obtained against Pseudomonas aeruginosa ATCC 27853 (11 mm) in ethyl acetate extract. However, the best antimicrobial activities were observed in hexane and methanol extracts for other tested microorganism. It was not determined any antimicrobial activity against Escherichia coli ATCC 25922. It has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. Ethyl acetate extract showed significant inhibitory value at the concentrations of 530 µg/ml (86.20 %) against PC-3 cell lines. Using methanol extract, the maximun inhibition was observed at concentration of 680 µg/ml with value of 51.86 %. The adsorbents were characterized via Scanning Electron Microscopy (SEM), Energy Dispersive Spectrometer (SEM-EDS), and Fourier transform infrared spectroscopy (FT/IR). Fomes fomentarius presented a low sorption capacity contrary to the two other fungi (RB and AA). RB and AA were selected as potential adsorbents for BR18 and RBBR, respectively. The maximum BR18 removal efficiencies for 10, 25, and 50 mg/L were obtained after 60 min to be 90, 88, and 86%, respectively. The RBBR adsorption efficiencies were 96.4, 96, and 90% for dye concentrations of 10, 25, and 50 mg/L. The adsorption of BR18 onto the RB biomass followed the Freundlich isotherm, while Langmuir is the best isotherm for RBBR sorption’s elucidation onto AA fungus biomass. The removal of BR18 by BR biomass was found to follow the pseudo-second order. In contrast, the adsorption of RBBR onto the AA biomass followed Lagergren’s pseudo-first order. For both adsorbents, the adsorption was exothermic, feasible, and spontaneous in nature. The major FAs of TL, TG, PL in both species were Cl6:0 (palmitic açid, PA), Cl8:1 n-9 (oleic acid, OLA) and Cl8:2 n-6 (linoleic acid, LA). In both species, total PUFA amounts were found to be higher than total MUFA and total SFA in TL, TG and PL fractions. The efficient production of the fatty acids especially linolenic and oleic acids which are majorly needed in building blocks of dietary human has confirmed these species as good source of nutrition. On the other hand, insufficiently studied cytotoxic activity (using pC-3 cell lines) of these mushrooms were investigated by using various solvent systems Ethyl acetate extract of Russula delica and, Pleurltus ostreatus showed significant inhibitory value at the concentrations of 520-530 µg/ml (99,45%- 92,82%) against PC-3 cell lines with IC50; 274,53-297,77 µg /mL respectively. Methanol extracts did not show any cytotoxic activity. lt has been found that the potential of cytotoxic activity is depended on concentration and solvent type of extracts. As a result, the present study is a guide for biochemiçal and nutritional values of the both species and can be useful for further investigation on plıarnıacological applications.Öğe Investigation of Nutritional Content, Antioxidant Anticancer and Antimicrobial Activities of Pleurotus eryngii (DC. Thenutritional contents of the P. eryngii were investigated in detail.Crude protein (g 100 kg-1), carbohydrate (g 100 kg-1), lipid (g 100 kg1), ash (g 100 kg-1), dietary fiber (g 100 kg-1) and energy value (cal100 g-1) of fruiting bodies were analysed. Fatty acid compositions oftotal lipid, phospholipid and triacylglycerol fractions of Pleurotuseryngii were investigated by Gas chromatography. Totally 13 differentfatty acids constituents were identified. Antioxidant propertiesincluding total antioxidant activity, free radical scavenging activity,reducing power activity, metal chelating activity, and superoxideanion radical scavenging activity were evaluated using different tests.The highest total antioxidant, DPPH scavenging, metal chelating, andsuperoxide anion radical scavenging activities were determined inethyl acetate extracts. The potential of cytotoxic activity on PC-3(prostate cancer) cell lines was found to depend on the concentrationand the type of the tested extracts fractions. It was determined thathexane, ethyl acetate, and methanol extracts of Pleurotus eryngii haveinhibition effect on the growth of tested microorganisms. The presentresults suggested that used as a daily nutrient, the Pleurotus eryngiicould be a novel source for new drug developments and promising insome cancer treatment.Öğe Preconcentrations of Pb(II), Ni(II) and Zn(II) by solid phase bio-extractor using thermophilic Bacillus subtilis loaded multiwalled carbon nanotube biosorbent(ScienceDirect, 2023) Ozdemir, Sadin; Dündar, Abdurrahman; Dizge, Nadir; Kılınç, Ersin; Balakrishnan, Deepanraj; Prasad, Kashi Sai; Senthilkumar, NatarajanAn alternative biotechnological solid phase bio-extraction (SPE) method was developed. Bacillus subtilis loaded multiwalled carbon nanotube was designed and used as biosorbent for the preconcentrations of Pb(II), Ni(II), and Zn(II). The experimental parameters such as sample flow rate, pH of sample solution, amounts of Bacillus subtilis and multiwalled carbon nanotube, volume of sample solution and reusability of column which affects the analytical characteristics of the SPE method were investigated in details. Surface structures were investigated by using FTIR, SEM. The best pH was determined as 5.0 and the percentages recoveries of Zn(II), Ni(II), and Pb(II) were determined as 99.1%, 98.7%, and 96.2%, respectively, at a flow rate of 3 mL/min. In this study, in which the profitable sample volume was determined as 400 mL and the amount of multiwalled carbon nanotube (MWCNT) as 50 mg. It was also observed that the column had a significant potential to preconcentrate Zn(II), Ni(II), and Pb(II) even after 25 reuses. The biosorption capacities for Pb(II), Ni(II) and Zn(II) were calculated as 34.2 mg/g, 36.8 mg/g and 45.5 mg/g respectively. The LOD values were calculated as 0.024 ng/mL for Pb(II), 0.029 ng/mL for Ni(II), and 0.019 ng/mL for Zn(II). The linear range was detected as 0.25–25 ng/mL. The concentrations of Pb(II), Ni(II), and Zn(II) in a variety of real food samples were determined by using developed method after application of certified reference sample.Öğe Several biological properties and synthesis of 2-(4-methylthiazol-5-yl) ethoxy substitute zinc phthalocyanine(John Wiley and Sons Ltd, 2023) Güngördü Solgun, Derya; Özdemir, Sadin; Dündar, Abdurrahman; Ağırtaş, Mehmet SalihIn this study, 4-(2-(4-methylthiazol-5-yl) ethoxy) phthalonitrile (3) and zinc phthalocyanine complex (4) were synthesized and characterized. UV–vis and fluorescence spectra, aggregation, and fluorescence parameters of the zinc phthalocyanine compound were investigated. As biological properties, DPPH radical capture, antidiabetic, DNA cutting, antimicrobial, photodynamic antimicrobial, anti-biofilm activities, and microbial cell viability parameters of the compounds were determined. The highest antioxidant activity was found as 62.80% at 100 mg/L concentration with compound 4, and compound 4 also showed the best antidiabetic activity as 65.17% at 400 mg/L concentration. In DNA cutting activity, it was determined that 3 and 4 cut DNA at all concentrations. For compounds 3–4, the minimum inhibitory concentration (MIC) values were found to be 32 and 16 mg/L for Enterococcus hirae and Enterococcus feacalis, respectively. The strongest MIC value of photodynamic antimicrobial activity was achieved as 4 mg/L for E. feacalis with 4. It was observed that both compounds inhibited the microbial viability activity of Escherichia coli by 100% at concentrations of 100 mg/L. Compound 4 inhibited biofilms of Pseudomonas aureginosa and Staphylococcus aureus as 92.48% and 98.56% at 50 mg/L concentration, respectively.Öğe Solid Phase Extraction Based on the Use of Agaricus arvensis as a Fungal Biomass for the Peconcentrations of Pb and Al Prior to Their Determination in Vegetables by ICP-OES(ATOMIC SPECTROSCOPY, 2013) Kilinc, E (Kilinc, Ersin); Dündar, Abdurrahman; Özdemir, Sadin; Okumuş, VeysiA solid phase extraction (SPE) method based on the use of a fungal biomass as sorbent was developed for the preconcentrations of Pb and Al. Agaricus arvensis was immobilized on Amberlite XAD-4 and loaded onto a column. Important parameters such as pH and flow rate of the solution, amount of biosorbent and resin, volume of sample solution, which affect the efficiency of the preconcentration procedure for Pb and Al, were optimized. The effect of possible interfering ions, i.e., Cd2+, Cu2+, Ni2+, Zn2+, Co2+, Na+, K+, Ca2+, Mg2+, and Fe2+, was investigated. Experiments on the repeatability of the solid phase column showed that the same column could be used at least 30 cycles without loss of the biosorption efficiency for the recovery of Pb and Al. The limit of detection (LOD) of Pb and Al was found to be 0.10 ng m(L-1) and 0.03 ng mL(-1), respectively. The sensitivity of ICP-OES improved 39.8-fold for Pb and 39.5-fold for Al. Linearity was obtained in the concentration range of 1.25-50.0 ng mL(-1) for Pb and 0.5-50.0 ng mL(-1) for Al. The relative standard deviation (RSD) of the method under optimum conditions was lower than 8.4% (n=5) for Pb and Al, which was validated through the analysis of certified reference tea and poplar leaves samples. The biosorption capacity of immobilized Agaricus arvensis for Pb and Al was found to be 31.2 mg g(-1) and 45.7 mg g(-1), respectively. The developed method was applied to determine the concentrations of Pb and Al in various edible vegetable samples (cucumber, okra, tomato, beans, aubergine, watermelon, zucchini, pepper, melon, lettuce, roka, purslane, scallion, cress, parsley, cabbage, sugar cane, and basil) grown along the cultivated banks of the Tigris River in Diyarbakir, Turkey.Öğe Synthesis and antioxidant, aggregation, and electronic properties of 6-tert-butyl-1,4-benzodioxine substituted phthalocyanines(Turkish Journal of Chemistry, 2018) Ağırtaş, Mehmet Salih; Cabir, Beyza; Gümüş, Selçuk; Özdemir, Sadin; Dündar, AbdurrahmanAs a starting material, 7-tert-butyldibenzo [b,e] [1,4] dioxine-2,3-dicarbonitrile was prepared by the reac- tion of 4-tert-butylcatechol with 4,5-dichlorophthalonitrile. Metallophthalocyanine complexes ( 4 { 7 ) were obtained by cyclotetramerization of 7-tert-butyldibenzo [b,e] [1,4] dioxine-2,3-dicarbonitrile. All compounds were characterized by elemental analysis and other spectroscopic methods (IR, UV/Vis, and 1 H NMR). Phthalocyanine compounds remained nonaggregated in tetrahydrofuran at the studied concentration ranges. Metallophthalocyanines ( 4 { 7 ) were tested for their antioxidant activities. The antioxidant activity processes included evaluation of radical-scavenging activity, chelat- ing activity, and reducing power. These compounds were compared to standard antioxidant ascorbic acid. The electronic data of the new compounds were obtained by computational calculations at the B3LYP/6-31G (d,p) level of theory.Öğe Synthesis and Characterization of Bionanomaterials and Evaluation of Their Antioxidant, Antibacterial, and DNA Cleavage Activities(Chemistry Select, 2021) Tarhan, Tuba; Dündar, Abdurrahman; Okumuş, Veysi; Çulha, MustafaIn this study, the hexagonal boron nitride (hBN) (1), poly-levodopa (P-L(DP) (2), P-L(DP) coated hBN (hBN@P(L-DP)) (3), and silver nanoparticles (AgNPs) decorated hBN@P(L-DP) (hBN@P(L-DP)-AgNPs) (4) were synthesized and characterized by multiple spectroscopic techniques. Additionally, their biological properties such as antioxidant, antibacterial, and DNA cleavage activities were determined. It is worth noting that, products 3 and 4 were newly synthesized structures. The antioxidant activities of all the nanomaterials 1, 2, 3, and 4 nanomaterials were investigated using some tests such as DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging and reducing power ability. Among the synthesized nanomaterials, product 2 exhibited the highest radical scavenging (64.8±1.94 %) and reducing power activity (0.61±0.017) at a concentration of 200 mg L−1. Product 4 was determined to have antibacterial activity against all three Gram-positive and three Gram-negative test bacteria. In addition, all the nanomaterials were tested for cleavage activity using pBR 322 plasmid DNA, as a result of which it was determined that only product 4 showed the ability of cleavage from both chains of DNA.Öğe Synthesis, characterization, aggregation behavior, antioxidant activity, and antibacterial activity of metallophthalocyanines carrying four phthalonitriles group(Editions de l'Academie Republique Populaire, 2016) Güven, M. Emin; Ağırtaş, M. Salih; Özdemir, Sadin; Dündar, AbdurrahmanA novel phthalonitrile derivative and zinc (II), cobalt (II), and cupper (II) phthalocyanines complexes were synthesis and characterized. The novel compounds have been characterized by using various spectroscopic data FTIR, UV/vis, NMR, elemental analysis etc. The aggregation investigations carried out in different concentrations indicate that 4- (4-(1-(4- phenoxyphenyl)-1-phenylethyl)phenoxy)phthalonitrile substituted phthalocyanine compounds do not have any aggregation behavior for the concentration range of 10-4-10-5 M in tetrahydrofuran. The antioxidant properties of the phthalonitrile and its phthalocyanine compounds were evaluated in three series of in vitro tests: DPPH free radical scavenging, ferrous ion chelating activity and reducing power. Antimicrobial activities of compounds were investigated.
